The birth of The University of Sydney in 1850 broke with the traditions of Britain’s ancient universities by admitting students on academic merit rather than on the basis of religion or social class. Fierce debate about The University of Sydney reform among academic and national leaders in Britain and North America had led to calls for tertiary education institutions to respond to social change and meet the demands of modern society.

The University of Sydney Act received royal assent on 1 October 1850. The University of Sydney emerged from a decade of "social and political revolution", as described by John Woolley, the University's first principal and professor of classics. The transportation of convicts to New South Wales had ended in 1840 while the arrival of free settlers had accelerated. Under pressure from activists such as William Charles Wentworth, there were moves towards self-government where the governor would no longer act autocratically, but only on the advice of the colony's elected representatives.

The first bursary at the The University of Sydney was the Maurice Alexander bursary, endowed in 1874 with a gift of £1000 from Alexander's widow. The bursary first supported Edward Raper, who quickly took advantage of the sporting opportunities at Sydney – he captained the University rugby football team in the late 1870s and led the first New South Wales team on its New Zealand tour in 1882. "This The University of Sydney is not only for those who have private means or professional connections to start them; it is founded for the people," said Badham in 1879, five years after the grant of the Alexander bursary. In the intervening years, bursaries had allowed 11 students to attend university who otherwise would never have had the opportunity.

The original Fisher Library, now the MacLaurin Hall, was purpose-built as an extension to the Quadrangle to house, The University of Sydney's increasingly large collection of books. Today's Fisher Library, built between 1958 and 1962, is one of the University's first modernist buildings.

In 1881 The University of Sydney decided to open its doors to women, admitting them on the same basis as men, among the earliest in the world to do so, and many decades before either the University of Oxbridge or Cambridge. The first generations of women to study at Sydney have left their mark in many ways. One of them, Jane Foss Russell, was in the second group of women to enrol at The University of Sydney, graduating with a BA in 1886 and an MA in 1889. Jane Foss Russell was passionate about higher education for women, and soon after graduation, went on a study tour of women’s colleges in England, including visits to Girton and Newnham colleges in England.

The University of Sydney (informally Sydney University or USyd) is a university located in Sydney, Australia. It is the oldest university in Australia, having been established in 1850. As of 2009, the university had 47,775 students, making it the second largest in Australia. The main campus is centered on large Oxbridge-inspired grounds which spread across the suburbs of Camperdown and Darlington on the south-western outskirts of the Sydney CBD. In addition, The University of Sydney encompasses a number of small special-purpose satellite campuses obtained from other organisations over the past 20 years, including the Sydney Conservatorium of Music and the Sydney College of the Arts. The University of Sydney is a member of Australia's Group of Eight, Academic Consortium 21, the Association of Pacific Rim Universities (APRU) and the Worldwide Universities Network.

The main campus is also the focus of The University of Sydney's student life, with the student-run University of Sydney Union (known as 'the Union') in possession of three buildings - Wentworth, Manning and Holme Buildings. These buildings house a large proportion of the university's catering outlets, and provide space for recreational rooms, bars and function centres. One of the largest activities organised by the Union is the Orientation Week (or 'O-week'), centring on stalls set up by clubs and societies on the Front Lawns.

The University of Sydney is currently undertaking a large capital works program with the aim of revitalising the campus and providing more office, teaching and student space. The program will see the amalgamation of the smaller science and technical libraries into a larger library, and the construction of a central administration and student services building along City Road. A new building for the School of Information Technologies opened in late 2006, and has been located on a site adjacent to the Seymour Centre. The busy Eastern Avenue thoroughfare has been transformed into a pedestrian plaza, and a new footbridge has been built over City Road. The new home for the Sydney Law School, located alongside Fisher Library on the site of the old Edgeworth David and Stephen Roberts buildings, has been completed.

The University of Sydney Library consists of thirteen individual libraries located across the university's various campuses. According to the library's publications, it is the largest academic library in the southern hemisphere: The University of Sydney statistics show that in 2007 the collection consisted of just under 5 million physical volumes and a further 300,000 e-books, for a total of approximately 5.3 million items. The Rare Books Library possesses several extremely rare items, including one of the two extant copies of the Gospel of Barnabas and a first edition of Isaac Newton's Philosophiae Naturalis Principia Mathematica.
